乙脑病毒CN株PrM/E基因的克隆及序列分析

Cloning and Sequencing of PrM/E Genes of Japanese Encephalitis Virus(JEV) CN Strain

摘要: 通过RT-PCR扩增乙型脑炎病毒CN株主要抗原基因PrM/E,并将PrM/E基因克隆、测序后,与GenbanK发表的JEVSA14(U14163)基因序列进行比较分析,发现CN株与JEVSA14强毒株的同源性为98%。在JEV基因组5'端477~2477的长2000nt的决定JEV抗原性的PrM/E区中,CN株与SA14株有40个碱基不同,其中28个碱基突变为同义突变,另外12个为错义突变,导致相应的氨基酸序列(666个)中的4个氨基酸发生了突变,其中有2个氨基酸出现在含有关键的抗原决定簇的E蛋白内,但不影响其功能表达

Abstract: This paper reports the cloning of the PrM/E gene of the Japanese Enceplialitis Virus(JEV) CN strain by RT-PCR and analysis of the gene sequence. Comparison of the gene sequence with that of the JEV SA14 strain demonstrates 98% sequence homology. In the 2 Kb genome region of the virus, between nucleotides 477~2477, 40 base pairs differe between the two strain. Only 12 of these differences are missense mutations that result in 4 amino acid differences in the protein sequences. Two of the amino acid substitutions occur in the region encoding for the E protein and do not cause a change in the activity of this protein between the two strains of JEV.
