石灰花楸的快繁技术研究

A Study on the Rapid Propagation Technique in Sorbus Folgneri

摘要: 以石灰花楸带芽茎段为试验材料,研究了石灰花楸组织培养和快速繁殖的适宜条件。试验表明,芽启动培养基为改良MS+2.0mg/L 6-BA+0.5mg/LNAA、在春季取材时,启动率达90%以上;最佳继代培养基为1/2MS+1.5mg/L 6-BA+10mg/LNAA,增殖系数大于4;生根培养基用添加1.0mg/LIBA+0.1mg/LNAA+0.5mg/L活性炭+10 g/L蔗糖的1/2MS时为宜,生根率达75%以上。

Abstract: By using stems with buds of Sorbus folgneri(Schneid.) Rehd as materials, the experiment was made to find out the best culture process and the proper culture medium. The technical system of tissue culture for rapid propagation was established. The results are as follows. The optimal initiation medium is MS+2.0 mg/L 6-BA+0.5 mg/L NAA, and when the stem is isolated in spring, the rate of initiation is over 90%. The optimal subculture medium is 1/2 MS+1.5 mg/L 6-BA+1.0 mg/L NAA, the multiplication rate of shoot clumps can surpass 4.00. The proper rooting medium is 1/2 MS+1.0 mg/L IBA+0.1 mg/L NAA+0.5 mg/L Ac+10 g/L sugar and the rooting rate can be up to 75%.
