2,4-D、KT对小麦成熟胚愈伤组织形成、分化的影响

Effect of 2, 4-D, KT on Callus Formation and Plantlet Regeneration from Mature Wheat(Triticum aestivum L.) Embryos

摘要: 以小麦新品种川农17、12成熟胚为外植体进行离体培养,结果表明:2mg/L 2,4-D有利于胚性愈伤组织形成,5mg/LKT有利于植株再生(两品种植株再生率分别为73.63%,89.47%)。高浓度2,4-D(4~8mg/L)虽不影响成愈率,但抑制植株再生。这种抑制作用能被KT拮抗。而KT的这种拮抗作用在2mg/L 2,4-D条件下不能体现。

Abstract: Mature wheat embryos of new wheat varieties Chuannong 17 and 12 were cultured in vitro. The results show that 2 mg/L 2, 4-D is beneficial to the formation of totipotent calli. 5 mg/L KT is beneficial to plant regeneration(The regeneration frequencies of the two cultivars are 73.63%, 89.74% respectively). High concentration of 2, 4-D(4~8 mg/L) has no bad effect on callus formation, but they inhibit plant regeneration. This kind of inhibition is resisted by KT, but in the initiation media supplemented with 2 mg/L 2, 4-D this kind of resistance disappears.
