盘基网柄菌(Distyostelium discoideum)基因芯片研制与基因表达检测

摘要: 将56个盘基网柄菌基因的PCR扩增产物制成基因芯片,提取不同发育阶段的总mRNA,逆转录制备cDNA探针,荧光染料标记,通过Southern杂交,检测盘基网柄菌不同发育阶段的基因表达模式。结果表明,在培养8h特异性表达的有actin、calmodulin、cdc2、Mo15等与孢子萌发和营养生长有关的基因;H2B-2和p24基因在16h时特异表达,说明这两个基因与盘基网柄菌由单细胞营养生长阶段向多细胞发育阶段的发育转化有关。

Abstract: Microarray was made with PCR amplification products of 56 Distyostelium discoideum genes. cDNA probes were prepared through reverse trnscription of total mRNA extracted at different developmental stages and tagged with fluorescent dye. Gene expression patterns at different developmental stages were detected through Southern hybridization. The result indicates that actin, calmodulin, cdc2 and Mo15 are related to spore germination and vegetative growth expressed specifically at 8 h of culture while H2B-2 and p24 expressed at 16 h of culture illuminates their relations to the transform from single cell stage of vegetative growth to multicellular stage of development.
