水稻光温敏核不育系花药培养的初步研究

A Study on Anther Culture of Photo period Temperature Sensitive Genic Male Sterile Line Rice

摘要: 利用光温敏核不育系材料蜀光637S、蜀光570S、蜀光582S,研究了花粉发育的不同时期、头季稻和再生稻、低温预处理时间、诱导培养基和分化培养基等5个方面对花粉愈伤组织的形成与绿苗再生的影响。结果表明:小孢子发育单核中,晚期的花药诱导效果优于其他时期;头季稻的愈伤及绿苗诱导率明显高于再生稻;低温预处理时间的延长有利于花粉诱导率的提高,处理8d左右达到最高诱导率;培养基SK3、NB对大多数材料的诱导效果优于其他培养基;增加IAA和少量的6-BA对提高分化率有益。

Abstract: By using PTGMS rice:SHU GUANG 637S, SHU GUANG 570S and SHU GUANG 528S as anther culture material, a study on different stage of microspore development, seedling rice and ratooning rice, low temperature pretreatment time, induction-media and differentiate-media was carried out. The result shows:the anthers with microspore at the mid- or late uninucleated stage are more effective to culture than that at the other stage. Anther derived from seedling rice is apparently higher than that from ratooning rice in both calluses inducted rate and green plantlet regenerated rate. Low-temperature pretreatment at 8~10℃ for 8 days is to stimulate anther induction. Medium SK3 and NB are better than the other media to most materials in callus induction. Adding IAA and 6-BA properly can improve the green plantlet differentiation.
