外源DNA导入水稻引起性状与分子变异初探

摘要: 利用浸渍法以萌动种子胚为受体导入籼稻不育系D汕A的总DNA,引起了常规稻蜀丰108的叶色、生育期和穗部性状的变异。同工酶及RFLP分析表明,外源DNA的导入引起了分子水平上的变异。

Abstract: The direct introduction of exogenous DNA without carrier into rice was carried out. The mature seed embryo of indica rice Shufeng 108 was soaked in the solution with 500μg DNA of the donor plant indica rice (D Shan A) per milliliter and 2% DMSO at 26℃ for 36 hours. The variantes were selected and their isozyme and RFLP analysis were carried out. The results are as follows:①exogenous DNA introduction results in variations, in which two plants show greener leaf color, bigger ears and later maturity than those of the control, and the variant traits are stable in the progeny; ②the variant plants are similar to control in isozyme patterns of esterase and peroxidase, but there are three and four more deep dyed enzyme bands than control, respectively, which displays the differences in enzyme activity; ③RFLP analysis indicates the DNA variances in variant plants.
