导入矮秆基因的小偃麦材料的细胞学和储藏蛋白研究

Cytological and Seed Storage Protein Analysis of Dwarf Lines with Wheat-Thinopyrum Intermedium Chromatin

摘要: 用小麦-中间偃麦草部分双二倍体与含Rht10的小麦品系杂交回交,培育了稳定的株高在50~70cm左右,农艺性状较优的矮杆小偃麦品系。我们对其中的2份材料进行了细胞学和醇溶蛋白分析。结果表明,材料攀89074-1-1-1-1、攀89074-3-2-1的染色体数分别为2n=56和2n=50。利用种子醇溶蛋白电泳分析表明它们与"中5"均具有反应偃麦草供体的特征带,在Gli-B1和Gli-D1上表现一定的差异,攀89074-3-2-1还含来自黑麦的1RS蛋白带,攀89074-1-1-1-1缺少"中5"的一条Gli-B1带。在高分子谷蛋白亚基上,攀89074-1-1-1和攀89071-3-2-1与"中5"在Glu-A1和Glu-B1上表现了较大的差异。结果认为,品系攀89074-1-1-1和攀89074-3-2-1是四川及南方麦区小麦育种中利用中间偃麦草基因的优良品种资源。

Abstract: Lines Pan 89074-1-1-1 and Pan 89074-3-2-1, with 5~70cm height,wee developed from a partial amphiploid wheat-Thinopyrum intermedium, line with Rht10 gene. Cytological analysis exhibits that the chromosome number of Pan 89074-1-1-1-1 and Pan 89074-3-2-1 are 56 and 50 respectively. APAGE of gliadin indicates that both Pan 89074-1-1-1-1and Pan 89074-3-2-1 containe band Gli-Agi1, but Pan 89074-1-1-1-1 is lack of band Gli-B1 than Zhong 5, and Pan 89074-3-2-1 has additional band of Gli-B1l, which shows that Pan 89074-3-2-1 possesses 1RS/1BL chromosome. The differences of HMW-GS between Pan 89074-1-1-1-1, Pan 89074-3-2-1 and Zhong 5 are obtained by SDS-PAGE analysis. The Agronomical traits also showed that lines Pan 89074-1-1-1-1 and Pan 89074-3-2-1 are novel resources for transfer Thinopyrum intermedium chromatin to wheat cultivars in south China.
