Pseudomonas属分离物对杉苗根附着的初步研究

摘要: 用选择性培养基从杉木幼苗根围分离细菌,发现根面的数量最多,比非根际土多达百倍;其中根面分离细菌中能离体拈抗立枯丝菌的分离物高达70%,而非根际土只只12.5%。利用紫外光诱变几个产生黄光的Pseudomonas分离物,能稳定地抗利福平,用以测定对杉木幼苗根面的附着,显示AP03-菌株随时间延长(5-50分钟)附着的细胞增加,并依赖于初始菌的密度,在每毫升含103-107个细胞范围内,紧密附着于根面的细菌数呈线性关系。在根的提取物中发现具有凝聚AP03-等菌株的凝聚因(但不凝聚大肠杆菌),推断是一种收白质。

Abstract: Isolates of fluorescent Pseudomonas were isolated from the seedling rhizoplane and rhizophere of China fir (Cunn-inghamia landeolata) With the fluorescent pseudomonad selec-tive media (Acc). The number of colonies from the rhizoplane was larger, and about 100 times more than that from the non-rhizosphere soil. the percentage of isolates, isolated from rhizoplane and in vitro antagonistic against Rhizoctonia solani on KB, reached 70%. the chosen strains, induced tolerant to rifampin by UV, Were used for binding assay. It wad found that the cells of AP03- adhesive to the root surface of China fir incrdased with time (from 5 to 60 minutes), and depended upon the starting concentration (form 103 to 107 cfu/ml) with a linear rdlation, the agglutinative factor in the root extracts was found out to agglutinate AP03-(not to E. coli), and was deduced as a protein.
