苹果褪绿叶斑病毒的抗血清制备及其检测应用研究

摘要: 用昆诺藜(Chenopodium quinoa)为诊断和分离寄主,从混合感染潜隐病毒的苹果花瓣中分离纯化出苹果褪绿叶斑病毒。经纯化的病毒,在繁殖寄主-昆诺藜上增殖扩大。采用聚乙二醇沉淀和差速离心技术,从感染病毒的昆诺藜中提纯出褪绿叶斑病毒作为抗原,免疫家兔,经心脏采血,制备出了该病毒的抗血清。用制备的抗血清进行了检测褪绿叶斑病毒的方法和对脱毒苗以及生产苹果树的带毒情况作了初步研究。A蛋白酶联免疫法适于检测苹果褪绿叶斑病毒。

Abstract: Chenopodium quinoa was used as isolate and propagation hosts of apple chlorotic leaf spot virus. CLSV was isolated from apple petales infected by apple latent viruses. Single lesion sub-selections were made and increased on C. quinoa. The virus was propagated on C. quinoa grown under control conditions. The antigen was purified by precipitation with polyethylene glycol M. W. 6000 and two cycle differen rential centrifugation at 4℃. Antiserum was used to detected CLSV in apple samples of CLSV-free and the virus trees from apple orchard. Two ELISA techniques(Indirect Method and Protein A ELISA) were compared. The Protein A ELISA method was suitable for detecting Chlorotie leaf spot virus in apple.
