分离LDH和酯酶同工酶的聚丙烯酰胺凝胶制备和LDH活性与反应时间关系的探讨

APPROACH TO THE CONCOCTION BETWEEN THE POLYACRYLAMIDE GEL DISCONNECTING LDH AND EST ISOENZYMES AND THE RELATIONSHIP OF TIME-ACTIVITY OF LDH ISOENZYM

摘要: 用正交试验方法,考察了聚丙烯酰胺凝胶电泳分离乳酸脱氢酶同工酶和酯酶同工酶的分离胶制备的三因素(丙烯酰胺浓度、丙烯酰胺/甲叉双丙烯酰胺、缓冲液PH)、三水平,即L9(33),采用综合评分法衡量电泳结果,经方差分析选出分离胶制备的最佳条件:乳酸脱氢同工酶,Acr=7.5%、C=2.6%,Acr/Bis=37.5、分离胶缓冲液PH8.9;酯酶同工酶,Acr=7.0%、C=2.3%。Acr/Bis=42.5分离胶缓冲液PH8.9。乳酸脱氢酶同工酶的活性与反应时间呈"S"型曲线关系。

Abstract: This paper deals with the method to seek for the optimum of the polyacrylamide gel disconnecting LDH and Est isoenzymes by the ortgogonal method. The three factors (Acr, Acr/Bis, Tris-HCl pH) and the three levels[L8(33)] of the polyacrylamicle gel were studied. The best conditions for the polyacrylamide gel disc electophoresis to disconnect the isoenzymes were Acr=7.5%, C=2.6%, Acr/Bis=37.5, Tris-HCl pH8.9 in the disconnecting LDH isoenzyme and Acr=7.0%,C=2.3%, Acr/Bis=42.5, Tris-HCl pH8.9 in the disconnecting Est isoenzyme respectively, which were selected by the synthetical Lscore and the variance analysis. The relationship of the time-activity of LDH isoenzyme was a "S" curve.
