香猪ATGL基因的克隆、验证及其在不同猪品种的编码区序列比较

Cloning and verification of ATGL gene in mini-pig and comparison of the coding sequence in different porcine breeds

摘要: ATGL基因是近年来发现的一个新基因,其表达产物脂肪甘油三酯脂肪酶(ATGL)被证明是催化三酰基甘油水解第一步反应的限速酶。本研究采用电子克隆和RT-PCR的方法克隆并验证了贵州小香猪ATGL基因的cDNA序列。克隆的香猪ATGL基因其cDNA序列全长2102 bp,其中编码区1461 bp,编码486个氨基酸。通过测序分析发现,不同猪品种间ATGL基因的cDNA序列及预测的蛋白质序列高度保守。

Abstract: ATGL gene was recently identified as a novel gene and its expression product was named as adipose triglyceride lipase (ATGL), and ATGL was proved to be the rate-limiting enzyme that performs the initial step in triacylglycerol hydrolysis. A total of 2102 bp cDNA sequence of ATGL gene was cloned by in silico cloning and reverse transcription-PCR verification from Guizhou mini-pig in this study. The sequence of ATGL gene from mini-pig was 1461 bp in the coding region and the speculated protein contains 486 amino acids. Sequencing analysis indicated that the cDNA sequence of ATGL gene and the speculated protein both conserved among different pig breeds.
