TLRs信号通路和促炎症细胞因子基因在鸡胚成纤维细胞感染新城疫病毒过程中的表达分析

The Induction of TLRs-mediated Antiviral Signaling Pathway and Inflammatory Cytokine in Chicken Embryo Fibroblasts Infected with Newcastle Disease Virus

摘要: [目的] 研究新城疫病毒对鸡胚成纤维细胞中TLRs抗病毒信号通路和促炎症相关细胞因子的诱导作用。[方法] 采用分离培养的原代鸡胚成纤维细胞感染中等毒力的新城疫病毒,并在感染后的4、8、12、16、24 h收集细胞,利用定量PCR检测TLRs信号通路及促炎症相关细胞因子在细胞新城疫病毒感染过程中的表达变化。[结果] 结果显示,TLR3、TLR7、TLR15,及其衔接蛋白基因MYD88、TRIF,以及下游基因IFN-α、IFN-β、Mx的表达量在病毒感染后呈现出显著上调(P<0.05)。同时促炎症细胞因子IL-1β、IL-6、TNF-α的表达量在病毒感染后也表现出显著上调(P<0.05)。[结论] 研究表明,在鸡胚成纤维细胞对新城疫病毒的识别过程中,由TLR3、TLR7、TLR15介导抗病毒信号通路,同时诱导产生促炎症细胞因子IL-1β、IL-6、TNF-α,并在新城疫病毒诱导的炎症反应过程中发挥重要的作用。

Abstract: [Objective] The purpose of this study is to investigate the induction of Toll-Like Receptors (TLRs)-mediated antiviral pathway and inflammatory cytokines in chicken embryo fibroblasts (CEFs)infected with Newcastle disease virus. [Method] We isolated CEFs for infection with NDV,and then analyzed the expression levels of TLR-mediated antiviral pathway and inflammatory cytokine genes in NDV-infected cell at 4,8,12,16,24 h.p.i (Hours-posted infection)using qRT-PCR. [Results] Results showed that the expression of TLR3,TLR7,TLR15 were significantly induced in NDV-infected CEFs (P<0.05),and significant increase were also observed in downstream genes MYD88,TRIF,IFN-α,IFN-β,Mx in NDV-infected chicken cells (P<0.05).In addition,we found that the expression levels of inflammatory cytokines,including IL-1β,IL-6,TNF-α,were also significantly up-regulated in NDV-infected chicken cell. [Conclusion] These results suggest that TLR3,TLR7,TLR15 play an important role in induction of IL-6,IL-1β and TNF-α to defend against Newcastle disease virus infection in chicken cells.
