Optimization of Cellulase Assisted Extraction Total Flavonoids from Broccoli Using Response Surface Method

摘要: 【目的】优化纤维素酶辅助法提取西兰花总黄酮工艺。【方法】在单因素实验基础上,选取酶用量、pH、酶解温度、酶解时间为自变量,总黄酮提取率为响应值,采用Box-Behnken实验设计方法,研究各自变量及其交互作用对总黄酮提取率的影响,并利用Design-Expert软件,建立了总黄酮提取率与提取过程中各因素的二次多项式模型。【结果】结果表明,纤维素酶辅助提取西兰花总黄酮的最佳工艺条件为:酶用量0.84%、pH 5.0、酶解温度50 ℃、酶解时间57 min,该条件下总黄酮提取率为1.251%,与预测值仅相差0.556%,验证了该模型的有效性,该工艺与传统提取方法相比,提取率提高了41%。【结论】该研究结果可为西兰花开发成天然抗氧化功能性产品提供理论基础和科学依据。

Abstract: 【Objective】 The aim of the study was to extract total flavonoids from Broccoli through optimizing cellulose process. 【Method】 Based on single-factor experiments, enzyme amount, pH, enzymolysis temperature and enzymolysis time were set as independent variables and the yield of total flavonoids was response value. Effects of each variable and their interactions on total flavonoids were determined using Box-Behnken method. The quadratic polynomial regression equations between total flavonoids and each variable were built using Design-Expert software. 【Results】 The results showed that the optimal extraction conditions of total flavonoids from Broccoli were: enzyme amount 0.84%, pH 5.0, enzymolysis temperature 50 ℃, and enzymolysis time 57 min. The yield of total flavonoids was 1.251% under the conditions. The relative error was 0.556% compared to predictive value, indicating the model fitted well with experimental data. In addition, the extraction yield was increased by 41% as compared to the traditional extraction method. 【Conclusion】 The results could provide a theoretical foundation and scientific basis for the development of natural antioxidant products of Broccoli.
