一粒系小麦高分子量谷蛋白亚基1Ay的多样性

Variations of High Molecular Weight Glutenin Subunit 1Ay in Einkorn Wheat

摘要: 通过十二烷基磺酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)对44份一粒系小麦1Ay亚基的检测结果表明,一粒系小麦中普遍存在表达的1Ay亚基。供试的乌拉尔图小麦、野生一粒小麦和栽培一粒小麦的分布频率依次为42.86%,81.25%和100%。同时,1Ay亚基的迁移率在物种间也存在明显多态性。3种供试物种中分离出齐平于和快于1Dy12,齐平、快于和慢于1Bx7,以及齐平于1By8共6种迁移率类型。其中,乌拉尔图小麦的1Ay全部在1Dy12亚基附近,而野生一粒小麦和栽培一粒小麦则以集中在1Bx7附近为主。尽管一粒系小麦1Ay的表达与地理来源有关,但也存在变异,体现在来源地不同的同一物种的1Ay表达频率相异,又体现在来自同一地区材料的1Ay迁移率存在明显多态性。一粒系小麦具有的较丰富的1Ay亚基变异类型,为小麦育种提供了资源。

Abstract: In this paper,the variations of high molecular weight glutenin subunit (HMW-GS) 1Ay in 44 lines of einkorn wheats were investigated by means of sodium dodecyl sulfate polyacrylamide-gel electrophoresis (SDS-PAGE).The expressed 1Ay widely existed in the species including T. urartu,T. boeoticum and T.moncoccum in einkorn wheat.The expressed frequencies of 1Ay in T. urartu,T. boeoticum and T. monococcum were 42.86%,81.25% and 100%,respectively.The polymorphism of 1Ay mobility in the tested species was observed.The six patterns,including the similar and faster subunits compared to 1Dy12,the similar,faster and slower subunits compared to 1Bx7,and the similar subunits to 1By8,were separated in the species.Among them,the mobilites of 1Ay from T. urartu were close to 1Dy12,while those from T. boeoticum and T. monococcum were close to 1Bx7.The distribution of 1Ay wasn't totally related to geographical origin.Within a species,there were variations on the frequencies of expressed 1Ay among different areas.There were variations on the mobilities among lines from same area.The diversity on 1Ay in einkorn wheat may be useful in wheat quality improvement.
