猪圆环病毒ORF2基因的插入对猪细小病毒VP2基因蛋白表达的影响

The Influence of the Insertion of ORF2 Gene of Porcine Circovirus on the Expression of VP2 Gene of Porcine Parvovirus

摘要: 将扩增得到的PCV2SC株ORF2基因插入PPVSC-1株VP2基因的HindⅢ(522bp处)和SacⅠ(1220bp处)2个特异限制酶切位点,并将此重组基因分别连接到含绿色荧光蛋白基因的伪狂犬病毒转移载体(pPI-2.EG-FP),脂质体转染系统转染COS-7细胞,倒置荧光显微镜观察重组基因表达情况。结合测序结果预测重组基因所编码的蛋白质的二级结构。

Abstract: Obtain PPV VP2 and PCV ORF2 fragments by PCR,then insert ORF2 into VP2 at HindⅢ (522 bp)and SacⅠ (1220 bp)position respectively and predict secondary structure of recombinant protein.Transfer the recombinant genes to eukaryotic expression vector pPI-2.EGFP to construct the recombinant plasmids pPI-2.EGFP.VO.H and pPI-2.EGFP.VO.S.Then the eukaryotic expression recombinant plasmids were transfected into COS-7 cells with Lipofectamine TM2000 and the recombinant genes were expressed and detected by fluorescence inverted microscope directly.Predict the secondary structure of recombinant protein.
