太湖猪成纤维细胞系的建立及其生物学特性研究

The Establishment of Fibroblast Cell Line and its Biological Characteristic Research in Taihu Pig

摘要: 以太湖猪皮肤为材料,采用组织块法培养成纤维细胞,并对其进行生物学特性检测,包括细胞形态观察、细胞冻存前和复苏后接种的存活率测定、生长曲线绘制、染色体核型分析。结果表明:所培养的成纤维细胞呈典型的成纤维细胞形态;细胞冻存前和复苏后接种的存活率分别为96.5%和93.2%;生长曲线呈"S"形,倍增时间为72h;对所制备的染色体片核型分析显示2n=38,XY。本次实验成功地建立了太湖猪成纤维细胞系,为在细胞水平上保存太湖猪种质资源提供了可能,并为其他研究提供了实验材料。

Abstract: Take Taihu pig as the study materials. A fibroblast cell line was successfully established by the method of explant adherence culture. Observations on morphology, determination of viability, dynamic growth, analysis of karyotype were carried out. The results showed that cell viability was 96.5% before freezing and 93.2% after thawing, Population doubling time (PDT) of cells was approximately 72 h, 2n=36, XY. The newly established cell line not only preserved the gemlplasm resources of this important Taihu pig on the cell level, but also provided valuable materials for the research of genome, postgenome and somacloning.
