植酸酶基因工程酵母PP-NPm-8培养条件的研究

Study on Culture Condition of Phytase Engineering Yeast PP-NPm-8

摘要: 对植酸酶基因工程菌PP-NPm-8的培养条件进行了研究,发现种龄、接种量、生长培养基初始pH值及生长时间、诱导培养基初始pH值、甲醇诱导浓度均对工程菌的产酶有很大影响,优化后的工程菌培养条件为:种龄16 h,接种量3%,麦芽汁生长培养基最适初始pH 4.5,生长培养时间18 h,麦芽汁诱导培养基最适初始pH值5.0,甲醇诱导终浓度4%。工程菌在此条件下诱导培养36 h后产酶量可达85067 U/mL,比未优化培养条件时提高了近一倍。

Abstract: The culture conditions of PP-NPm-8 strain were studied in this paper. The results show that:the seed age is 16 h, the inoculation rate is 3%, the initial pH in growth phase is 4.5, and the growth phase is 18 h. However, in induction phase, the initial pH of malt wort is 5.0, and inducing concentration of methanol is 4%. Under the optimal culture conditions, its yield of phytase can come to 85067 U/mL which is about 1 times higher than that of the control.
