大熊猫皮肤组织酶解分离效果研究

Studies on Enzyme Digestion of Cutaneous Tissue of Giant Panda(Ailuropoda Melanoleuca)

摘要: 大熊猫皮肤组织块在200IU/mL胶原蛋白酶Ⅰ和0.25%胰蛋白酶中,分别在37℃、4℃下进行酶解的4个方案中,以在200IU/mL胶原蛋白酶Ⅰ,在4℃下酶解48~72h的酶解效果最好,活细胞的得率达到4.604×106/g,且胶原蛋白酶Ⅰ酶解得到的细胞以成纤维细胞为主。

Abstract: In the paper, we studied that custaneous tissue of giant panda was digested with collagenase-Ⅰ(200 IU/mL) or trypsin(0.25%) under 37℃ or 4℃. In the 4 treatments, the effect shows:the method of enzyme digestion which was treated with collagenase-Ⅰ(200 IU/mL) under 4℃ for 48~72 h was the best in all methods and the rate of living cells reached 4.604×106/g. In addition, the fibroblast cells was main cells in the test treated with collagenase-Ⅰ(200 IU/mL).
