双孢蘑菇菌丝体酯酶同工酶及可溶性蛋白的电泳分析

Studies on the Esterase Isozyme and Soluble Protein Electrophoresis of 15 Different Agaricus bisporus

摘要: 对引进的15株双孢蘑菇菌丝体进行了酯酶同工酶和可溶性蛋白的聚丙烯酰胺凝胶电泳分析。结果表明:15株双孢蘑菇菌丝体在酯酶同工酶以及可溶性蛋白质方面都表现出了一定的遗传性差异。进一步通过UPGMA平均连接程序进行聚类分析发现,15株双孢蘑菇在酯酶同工酶上,当相似水平为0.63时聚为一类;在可溶性蛋白方面,当相似水平等于0.94时被聚为一类。结果说明酯酶同工酶和可溶性蛋白电泳图谱对于双孢蘑菇种级水平上的分类具有重要作用,两者中尤以酯酶同工酶酶谱最佳。

Abstract: The esterase isozyme and soluble protein of 15 different A. bisporus strains were analysed with polyacrylamide gel electrophoresis. Both the esterase isozyme and soluble protein electrophoresis indicated genetic difference among 15 different A. bisporus. Then the genetic similarity was analysed by UPGMA based on the esterase isozyme and soluble protein gel electrophoresis. The result shows that according to the esterase isozyme gel electrophoresis, all strains collect together at the level of 77%;and according to soluble protein electrophoresis, all strains collect together at the level of 94%. All the results show that electrophoresis analysis of esterase isozyme and soluble protein, especially esterase isozyme, is a useful supplementary means for the classification of A. bisporus at the level of species.
