小麦新品系98-1266与川麦28在分子水平上的差异性研究

The Molecular Genetic Differences between Advanced Wheat Line 98-1266 and Chuanmai 28

摘要: 应用随机扩增多态性DNA(RAPD)和微卫星(SSR)标记,对小麦优质兼抗丰产1BL/1RS易位系98-1266与川麦28号在DNA分子水平上的遗传差异进行了检测。结果表明,在所用127个RAPD随机引物中,有4个引物能稳定地揭示材料间的差异性。该4个引物共扩增出17条带,其中7条带(占35%)具有多态性。在24个SSR位点上,共检测到34个等位基因。其中,有8个位点(占33.3%)能揭示98-1266与川麦28号间不同基因型的差异。本文还对RAPD和SSR标记在小麦遗传分析中的应用进行了探讨。

Abstract: By using RAPD and SSR markers, the genetic variations between 98-1266 and Chuanmai 28 were detected. 98-1266 is a new 1BL/1RS translocation line conferring excellent quality, resistance to powdery mildew and stripe rust, and high yield. Among 127 RAPD primers, 4 primers could amplify reproducible polymorphic bands between 98-1266 and Chuanmai 28. Seventeen bands are amplified from the 4 RAPD primers, among which 7 bands(35%) are polymorphic. In SSR analysis, eight out of 24(33.3%) SSR loci are polymorphic. On 24 SSR loci, a total of 34 alleles are detected in 98-1266 and Chuanmai 28. The applications of RAPD and SSR markers in wheat genetic research are also discussed in this paper.
