玉米胚性愈伤组织经不同方法导入大豆DNA形成的幼苗同工酶分析

摘要: 以玉米胚性愈伤组织为受体,以大豆DNA为供体,采用超声波法、减压渗透法和电激法转化玉米,所获后代材料采用PAGE电泳法对其进行酯酶及过氧化物酶同工酶分析。结果显示:多数受试材料与对照材料之间其酶谱差异明显,表明导入处理已对受体基因组产生较大影响;各转导方法的转化效果依次为:超声波法> 电激法> 减压渗透法。

Abstract: With ciecompress? osmosis,ultrasonication and electorpovation,the DNA of soybean was transmitted to embryogenic calli of maize.The peroxidase and esterase isozyme derived from the progenies of maize by polyacrylamide gel electrophoresis were analysed.The result shows that the isozyme patterns between the transformed and control plants presented distinct difference;the transmission had great effect on the genome of the host s.Conversion efficiencies of the different methods were compared in the experiment,and the order from the highest to the lowest was:ultrasonication,electroporation,ciecompress osmosis.
