来航鸡门静脉血浆中寡肽的高效液相色谱法分析

DETERMINATION OF OLIGO PEPTIDES IN PORTAL PLASMA OF THE LEGHORN BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY (HPLC)

摘要: 用Spherisorb C18反相柱,以0.1%三氟乙酸-水和0.1%三氟乙酸-乙腈作流动相,紫外光检测波长225nm,在PICO-TAGTM氨基酸分析仪上对来航公鸡门静脉血浆中的寡肽进行分析,得线性方程式y=5.4214×10-6x-0.0285,相关系数r=0.9977(n=5),变异系数cv%=2.856%(n=5),分析时间35min,采用保留时间定性,外标法以峰面积为基础定量,回收率平均为95.76%(n=3)。

Abstract: In this paper, the oligo peptides in portal plasma of the leghorns were analysed by high performance liquid chromatography (Waters PICO-TAGTM Amino Acids Analyser) with spherisorb C18 reverse-phase column (5μm, 5mm×250mm). The mobile phase A is 0.1% Trifluoressigsaure acid (TFA) in pure water. The moile phase B is 0.1% TFA in acetonitrile, and the ultraviolet wavelength is 225nm, and the running time is 35 minutes. The result is qualitative analysis based on the retention time (Rt). By measuring peak area, comparing it wity external standards and taking Gly-Tyr as example, the following results are reached the regression equation:y=5.2414×10-6x-0.0285, r=0.9977 (n=5), cv%=2.856% (n=5), the average rates of recovery of Gly-Tyr:95.76% (n=3).
