测定摩杂一代水牛毛中孕酮含量作早孕诊断的研究

DETERMiNATION OF HAIR PROGESTERONE CONTENT IN BUFFALOES FOR EARLY PREGNANCY DIAGNOSIS

摘要: 以毛为样品,应用竞争性蛋白质结合分析法测定孕酮含量,进行摩杂一代水牛早孕诊断试验,结果表明:人工授精后14天以前,孕与未孕牛的毛孕酮含量差异不显著(P> 0.05),16-18天差异显著(P<0.05),20-24天差异极显著(P<0.01)。据此,确定配种后24天的毛样判断妊娠与未妊娠的孕酮标准是:大于4.5毫微克/100毫克毛为妊酮,小于3.5毫微克/100毫克毛为未妊娠,介于两者之间为可疑。孕与未孕的诊断准确率分别为88.9%和100%。

Abstract: Preliminary study on early pregnancy diagnosis in 10 buffaloes (Murrah x Local swamp buff, F1) based on hair progesterone determination by means of a competitive protein-binding assay has been conducted. Measurements of progesterone profile during oestrus cycle showed that the difference in hair progesterone content between pregnant and non-pregnant buffaloes became significant (p<0.05) and highly significant (p<0.01) during days 16-18 and 20-24 post-oestrus respectively. When hair samples were taken at the back of 17 buffalloes and on the 24th day after oestrus, the accuracy of pregnancy diagnosis was 88.8% and 100% for pregnant and non-pregnant animals respectively as compared with the results obtained from rectal palpation two months after insemination, the criterion for discriminating pregnancy and non-pregnancy was taken as that:higher than 4.5ng/100mg hair, pregnancy, lower than 3.5ng/100mg hair, non-pregnancy.
