大花蕙兰组培苗耐寒性的综合评价

Comprehensive Evaluation on Cold Resistance of Cymbidium hybridum Varieties

摘要: 【目的】比较‘黄金岁月’(Cymbidium Lovely Moon 'Crescent')、‘红霞’(Cymbidium Royal Red 'Princess Nobuko')和‘晨辉’(Cymbidium Twilight Moon 'Daylight')3个品种大花蕙兰耐寒性的强弱,筛选大花蕙兰耐寒性鉴定的生理指标。【方法】采用人工模拟低温的方法,设置10、5、0和-5 ℃共4个低温处理,探讨其对3个品种大花蕙兰地上部和地下部丙二醛(MDA)、膜透性(CMP)、可溶性蛋白质(SP)、可溶性糖(SS)、超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)等7项生理指标的影响,利用主成分分析、隶属函数法和逐步回归分析,对这7个耐寒性生理指标进行综合评价。【结果】3个不同品种大花蕙兰在受到低温胁迫时,3种酶活性有不同程度升高,其中SOD和POD活性增加幅度最为明显,其耐寒性强弱的排序为:‘黄金岁月’>‘红霞’>‘晨辉’。【结论】从不同生理指标的变化趋势来看,细胞膜透性、SOD和POD活性的变化趋势可反映出不同品种的大花蕙兰组培苗的耐寒性的强弱,因此,可将细胞膜通透性、SOD和POD活性作为其耐寒性的鉴定指标。地上部和地下部在反映组培苗耐寒性能方面存在差异性。

Abstract: 【Objective】 The aim of the study was to compare cold resistance among three varieties of C.hybridum and to select physiological indicators of cold-resistance of C.Hybridum. 【Method】 This study included four low temperature treatments (10, 5, 0 and -5 ℃) by artificial simulation method. The effects of low temperatures on malondialdehyde (MDA), Membrane permeability (CMP), soluble protein (SP), soluble sugar (SS), superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities of aboveground and underground parts of three C.hybridum varieties were investigated. The seven cold-resistance physiological indexes were comprehensively evaluated using principal component analysis, membership function analysis and stepwise regression analysis. 【Results】 Three C.hybridum when subjected to cold stress, there are three kinds of activity increased to varying degrees, of which the magnitude of SOD and POD activity increased the most obvious, the order of cold-resistance capacity of varieties was Lovely C. Moon 'Crescent' > C.Royal Red 'Princess Nobuko' > C.Twilight Moon 'Daylight'. 【Conclusion】 CMP, SOD and POD activities were significantly associated with cold resistance, and thought be the indexes to identify cold resistance in C.hybridum. There were differences in cold-resistance between aboveground and underground components.
