猪载脂蛋白E基因的组织表达谱分析

Analysis of tissue-specific expression profile of porcine APOE

摘要:

载脂蛋白E在胆固醇和其他脂质的代谢中起着至关重要的作用,对猪APOE基因的组织表达谱分析是研究该基因功能的重要基础工作。本研究采用RT-PCR的方法对APOE在长白猪的13个组织中的表达进行了研究,结果显示APOE基因主要在肝和脑组织表达,其它外周组织如脾、肺、肾、大肠、小肠也有不同程度的表达,而在心、胃、背肌和淋巴中几乎不表达。本试验通过对猪APOE的组织表达谱分析,旨在为下一步研究猪APOE基因的功能奠定良好基础。进化上的保守性和功能上的重要性提示以猪为动物模型研究与APOE基因相关的人类疾病如高血压、心血管疾病、动脉粥样硬化、老年痴呆症等具有重要的科学和现实意义。

Abstract:

Apolipoprotein E plays an important role in cholesterol and other lipid metabolism and study on tissue expression files of porcine APOE gene is an important basis for researching functions of the gene. In this study, RT-PCR method was used to study on expression of APOE in thirteen tissues of Landrace and the results showed that APOE gene were mainly expressed in the liver and brain tissue, and there were different expression in other peripheral tissues such as spleen, lung, kidney, large intestine, small intestine, whereas almost no expression in heart, stomach, M. longissimus and lymph. The ultimate goal is to lay a good foundation for further studying the function of porcine APOE. It was suggested that pig is an ideal animal model for researching human diseases associated with APOE gene such as hypertension, cardiovascular disease, atherosclerosis, and senile dementia based on evolutionary conservation and functional importance of APOE.
