血清5型和13型副猪嗜血杆菌二价灭活苗的制备和免疫效力研究

Preparation and Immune Efficacy of prepared bivalent inactivated vaccine of serovar 5 and serovar 13 against Haemophilus parasuis

摘要: 摘要: 本研究旨在制备一种用于预防副猪嗜血杆菌病的二价灭活苗,并对其免疫效力进行评价。选择四川地区流行的副猪嗜血杆菌(Haemophilus parasuis,,HPs)血清5型MC-203株和血清13型XJ-103株,用甲醛灭活后,经PBS浓缩,将2种菌液与佐剂(铝胶)按1:1:2的比例混合制出二价灭活苗。随后用制备的灭活苗免疫仔猪进行相关安全性检验,抗体滴度检测。同时为了对比制备疫苗与国产商品疫苗和进口商品疫苗的免疫效果,笔者进行了仔猪免疫保护效果评价实验。结果如下:①用0.2%的甲醛在37℃作用 20 h可将HPs完全灭活。②灭活苗免疫仔猪后未造成不良反应,安全性好。③免疫仔猪后14 d即可在体内检测到HPs特异性抗体,196 d的血清中仍可检测到HPs抗体。④仔猪免疫保护效力试验表明,本实验所制备的灭活疫苗、进口商品疫苗和国产商品疫苗的免疫保护率分别为80%、80%、60%。以上结果表明,本研究制备的HPs灭活苗免疫仔猪,可以产生较好的免疫应答,并且能在一定程度上抵御HPs的感染。

Abstract: Abstract: This study developed a new kind of bivalent inactivated vaccine aiming at controlling Haemophilus parasuis (HPs) disease, and the vaccine’s immune effect was evaluated. The most prevalent serotype 5 strain MC203 and serotype 13 strain XJ103 in Sichuan province are selected, which was followed by inactivated with 0.2% formaldehyde, and then washed with PBS, after which, the MC-203, XJ-103 and alhydrogel adjuvant were mixed with a scale of 1:1:2 to make HPs bivalent vaccine. The specific antibody against HPs were detected through ELISA and safety evaluated in animal challenge experiments. The antibody titer of the bivalent inactivated vaccine against HPs were evaluated in piglets. In order to compare the immune efficacy among the bivalent inactivated vaccine, imported inactivated vaccine and domestic inactivated vaccine, piglets were immunized with the three kinds of vaccines and the immune efficacy was evaluated by results of immunized protection. The results were as follows: ①HPs was inactivated completely with 0.2% formaldehyde after incubation for 16 h at 37℃. ②HPs inactivated vaccine has a high security in piglets and without undesirable reaction. ③The specific antibody against HPs could be detected at the 14 day and lasted at least for 196 days after vaccination. ④Comparison of the immunity effectiveness of the inactivated vaccine demonstrated that the inactivated vaccines, imported inactivated vaccine and domestic inactivated vaccine immune have a protection of 80%, 80% and 60%, respectively. Through above, we got the conclusion that inactivated HPs vaccine in this study has an expected protection in piglets against predominant serotype of HPs.
