猪乙型脑炎病毒基因I型弱毒株免疫小鼠后诱导INF-γ的能力

The ability of genotype I swine Japanese encephalitislive- attenuate strain to induce INF-gamma in mice after immunization

摘要: [目的]评估基因I型猪乙型脑炎病毒弱毒株(SCYA201201株)作为候选疫苗诱发机体产生细胞免疫的能力。[方法]将基因I型猪乙型脑炎弱毒株(SCYA201201株F122代次)腹腔免疫小鼠后使用ELISA检测试剂盒测定早期的INF-γ水平,之后测定INF-γ在小鼠体内的持续时间。[结果]结果显示:猪乙型脑炎SCYA201201弱毒株(F122代次)经腹腔免疫小鼠后,小鼠血清中的INF-γ在免疫后6h就可上升到一个较高水平,且INF-γ在血清和小鼠脾脏淋巴细胞中均可持续14d以上。[结论]猪乙型脑炎SCYA201201弱毒株(F122代次)免疫小鼠后能够在早期诱发INF-γ产生,并且能够维持14天以上。有作为候选疫苗的潜力。

Abstract: [Objective] To evaluate Japanese encephalitis virus SCYA201201 strain of the genotype I induce cell immunity as vaccines candidate in mouse model. [Method] This research using ELISA kit the INF-γ detect the INF-γ levels of mice serum after immune with genotype I of Japanese encephalitis live-attenuated strain (SCYA201201 strain). [Results] The results showed that Japanese encephalitis live-attenuated SCYA201201 strain (F122 generation) of mice can induced INF- gamma level to a higher level when post immunization 6h in mice serum, and INF- gamma in serum and lymphocytes were sustained for more than 14days. [Conclusion] The study proved that the live-attenuated SCYA201201 strain can induce the production of INF- gamma in early immunity stage, and it can be maintained for a long time. There is potential for a candidate vaccine。
