响应面法优化产内切葡聚糖酶重组大肠杆菌发酵培养基

Optimizaiton of Culture Medium of Recombinant Escherichia Coli for Endoglucanase Production by Response Surface Methodology

摘要: 【目的】对产内切葡聚糖酶重组大肠杆菌的发酵培养基进行优化。【方法】采用单因素实验和响应面 Box-Behnken 实验设计相结合,对培养基的碳源及浓度、氮源及浓度和无机盐离子及浓度进行优化。【结果】结果表明,最佳的碳源、氮源和无机盐离子及添加量分别为乳糖0.63%,酵母粉1.06%,氯化镁0.11%,发酵产酶酶活最大值达到191.23 U/mL,比优化前52.43 U/mL提高了3.65倍。【结论】利用响应面法Box-Behnken实验设计对产内切葡聚糖酶的重组大肠杆菌K369R进行培养基发酵条件优化,为内切葡聚糖酶在工业中的大规模生产提供理论依据。

Abstract: 【Objective】The study aimed to optimize the fermentation medium of recombinant E. coli for endoglucanase production.【Method】Single factor experiment and response surface Box-Behnken design were used to optimize the carbon source and concentration, nitrogen source and concentration of the medium, and the concentration of inorganic salt.【Results】The result showed that the best carbon source, nitrogen source and inorganic salt ion were lactose, yeast powder and magnesium chloride respectively. The addition amount of various factor in fermentation medium was 0.63% lactose, 1.06% yeast powder, 0.11% magnesium chloride.Under the optimized conditions, the maximum endoglucanase activity reached 191.23 U/mL, which was 3.6 times higher than that before optimization of 52.43 U/mL.【Conclusion】The fermentation conditions of recombinant Escherichia coli K369R were studied by using the response surface method Box-Behnken experiment. This research can provide the basis for the endoglucanase manufacture.
