阿月浑子炭疽病菌的种类鉴定

Species Identification of Pathogen Causing Pistacia vera L. Anthracnose

摘要: [目的] 对阿月浑子炭疽病在河北省的危害、症状特点以及病原菌的种类进行研究。[方法] 2005-2008年分别从河北省涉县和唐县两个病区采集发病材料,进行病菌分离,对获得的分离菌株通过显微观察、致病性测定、寄主范围测定、生物学测定以及rDNA-ITS序列同源性比较进行病菌的种类鉴定。[结果] 所获菌株对阿月浑子均可致病,并可引起草莓叶片发病,但对辣椒、葡萄、黄杨的叶片以及苹果的果实均不致病。以菌株CN504的DNA为模板,扩增得到了全长为583 bp的DNA片段,并获得了该菌的rDNA-ITS序列。将该序列与GenBank中已有的DNA序列进行同源性比较,发现与其同源性最高的100个ITS序列菌株均为胶孢炭疽菌Colletotrichum gloeosporioides Penz或胶孢炭疽菌的有性态围小丛壳菌Glomerella cingulata Stonem,CN504与它们的同源性在98%~99%之间。[结论] 根据病原菌的形态和生物学特征,以及ITS序列同源性比较的结果,将阿月浑子炭疽病的病原菌鉴定为胶孢炭疽菌C. gloeosporioides

Abstract: [Objective] The aim of the study was to investigate the symptom characteristics and pathogen types of Pistacia vera anthracnose in Hebei. [Method] The strains were collected from Shexian and Tangxian counties in Hebei province from 2005 to 2008 and all isolates showed similar characteristics in colony.Pathogen was identified by microscopy,pathogenicity,host range,biological analysis and homology comparison of rDNA-ITS sequences. [Results] All isolates infected strawberry leaves but did not infect pepper,grape,Japanese euonymus leaves and apple fruits.The DNA of strain CN504 was used as a template,and one rDNA-ITS fragment of 583 bp was amplified.The obtained sequences were compared with those in GenBank.One hundred sequences with the highest homology belonged to Colletotrichum gloeosporioides Penz or sexual stage of Glomerella cingulata Stonem.Their homology ranged from 98% to 99%. [Conclusion] Based on morphology,biological traits and sequence analysis of internal transcribed spacer region of ribosomal DNA (ITS),pathogen of pistachio anthracnose was identified as C. gloeosporioides.
