异源表达AtCIPK23基因对马铃薯试管薯结薯及耐低钾抗性的影响

Effects of AtCIPK23 Gene Ectopic Expression on vitro Tuberization and Tolerance against Low-K~+Stress in Potato

摘要: [目的] 研究低钾处理下异源表达AtCIPK23基因对马铃薯试管薯结薯的影响。[方法] 以马铃薯品种鄂薯3号(W)及其转AtCIPK23基因的株系L1、L23和L34为材料,通过试管薯诱导体系,研究马铃薯试管薯在不同钾浓度处理下的单瓶结薯数、薯块直径以及植株和试管薯的钾含量,试管薯淀粉与蔗糖的含量。[结果] 2mmol/L和10mmol/L处理下,L1、L23和L34的单瓶结薯数、薯块直径均高于对照W,且植株和试管薯的钾含量,试管薯淀粉与蔗糖的含量均高于对照W。正常钾(20mmol/L)处理下,对照与转基因各株系间各指标的差异不显著。[结论] 异源表达AtCIPK23基因提高了马铃薯试管薯对低钾的抗性。

Abstract: [Objective] The purpose of this study was to research effects of ectopic expression AtCIPK23 gene on potato tuberization in vitro under low potassium treatments.[Method] Three transgenic potato lines (L1,L23 and L34) and non-transgenic control W (E3 cultivar) were used to explore tuber growth in low K+ condition by microtuber induction system.The number of tubers in single bottle,diameter of potato,potassium content in potato plant and microtuber,the content of microtuber starch and sucrose were determined under different concentrations of potassium.[Results] The number and diameter of mirotuber in transgenic lines L1,L23 and L34 were higher than that in W under 2mmol/L and 10mmol/L potassium concentration treatments.Potassium content in plant and microtuber,sucrose and the starch content in microtuber of transgenic lines were higher than that of non-transgenic lines in these potassium concentrations.In 20mmol/L potassium concentration,these indexes between transgenic lines and control were no-significant different.[Conclusion] AtCIPK23 gene ectopic expression in potato could increase the resistance to low potassium condition.
