引种紫锥菊的ISSR和SRAP遗传多样性分析

The Analysis of Genetic Diversity of Introduced Echinacea purpurea Moench Based on ISSR and SRAP

摘要: [目的] 对不同来源的引种紫锥菊进行遗传多样性分析。[方法] 采用ISSR和SRAP分子标记技术,分析14份引种紫锥菊材料的多态性条带比率,计算各品种间的遗传相似系数,建立各品种间的聚类图。[结果] 8个ISSR引物和9个SRAP引物对分别扩增出105和171条带,多态性比率分别为81.9%和80.7%;两个标记综合分析表明紫锥菊样品间的亲缘关系与种质来源地之间存在一定相关性。[结论] ISSR和SRAP可以应用于紫锥菊种内遗传多样性分析,其综合分析结果优于单独分析;紫锥菊种内不同来源样品材料间存在丰富的遗传多样性。

Abstract: [Objective] The objective of this study was to detect the genetic diversity among 14 Echinacea purpurea moench introduced from different germplasm source. [Method] Inter-simple sequence repeat(ISSR) and sequence-related amplified polymorphism(SRAP) molecular markers were used in this case. [Results] 105 and 171 bands were obtained by ISSR and SRAP markers amplified through 8 and 9 selected primers respectively. The PPB(percentage of polymorphic bands) was 81.9% in ISSR and 80.7% in SRAP. [Conclusion] Combining with two methods would be more suitable for the analysis of genetic diversity, and its results of comprehensive analysis are better than that of separate analysis. There was rich genetic diversity among these Echinacea purpurea moench.
