香猪ATGL基因的克隆、验证及其在不同猪品种的编码区序列比较

Cloning and Verification of ATGL Gene in Mini-pig and Comparison of the Coding Sequence in Different Porcine Breeds

摘要: ATGL基因是近年来发现的一个新基因,其表达产物脂肪甘油三酯脂肪酶(ATGL)被证明是催化三酰基甘油水解第一步反应的限速酶。本研究采用电子克隆和RT-PCR的方法克隆并验证了贵州小香猪ATGL基因的cD-NA序列。克隆的香猪ATGL基因其cDNA序列全长2 102 bp,其中编码区1 461 bp,编码486个氨基酸。通过测序分析发现,不同猪品种间ATGL基因的cDNA序列及预测的蛋白质序列高度保守。

Abstract: ATGL gene was recently identified as a novel gene and its expression product was named as adipose triglyceride lipase(ATGL), which was proved to be the rate-limiting enzyme that catalyse the initial step in triacylglycerol hydrolysis.A total of 2 102 bp cDNA sequence of ATGL gene was cloned by using the electronic cloning and RT-PCR methods from Guizhou mini-pig in this study.The coding region of ATGL gene cDNA was 1 461 bp, which encoded 486 amino acids.Sequencing analysis indicated that the cDNA sequence of ATGL gene and the predicted protein sequences were highly conserved among different porcine breeds.
