黑麦属NBS-LRR类抗病基因同源序列的分离与鉴定

Isolation and Identification of NBS-LRR Resistance Gene Analongs from Secale L.

摘要: 根据已知植物抗病基因(resistance gene,R gene)编码的蛋白质NBS-LRR保守结构设计了10对特异简并引物,对5份黑麦材料的基因组DNA进行抗病基因同源序列克隆,共获得45条抗病基因同源序列(resistance geneanalogs,RGAs)。同源性比较发现,RGAs序列之间的核苷酸相似性是41.9%~99.6%,其中16条具有连续开放阅读框(ORFs)。同时,利用MEGA 3.1将这16条序列与4个已知R基因进行聚类分析,发现有13条序列与RPM1亲缘关系近,2条与Pm3a亲缘关系近。本研究在黑麦中获得的RGAs既可用于筛选黑麦的抗病候选基因,同时也可以作为分子标记,用于作物分子辅助选择育种,且对进一步研究黑麦中NBS-LRR类R基因的起源和遗传进化提供参考和依据。

Abstract: The resistance gene analogs(RGAs) from 5 Secale L.accessions were amplified using 10 pairs of specific degenerated primers based on the conserved regions of nucleotide binding site-leucine rich repeat(NBS-LRR) domain of plant resistance genes(R genes).A total of 45 RGAs were isolated, the nucleotide sequences of the 45 RGAs showed the similarity of 41.9% to 99.6%.Sixteen out of 45 RGAs had the uninterrupted open reading frames(ORFs).Thirteen out of 16 RGAs with RPM1 had near relation, two RGAs were close to Pm3a.These results can be used for the isolation of related resistance genes from Secale, and the development of molecular markers for the marker-assisted selection(MAS) of crop breeding programs.Furthermore, it can provide an evidence for the studies on the origin and genetic evolution of NBS-LRR type R gene of Secale in the future.
