球孢白僵菌原生质体制备和再生条件

Preparation and Regeneration of Beauveria bassiana(Balsamo) Vuillemin Proto

摘要: 球孢白僵菌[Beauveria bassiana(Balsamo) Vuillemin]经液体培养后,采用酶解方法制备其菌丝原生质体,并采取单因素实验对再生条件进行了研究。结果表明,经纤维素酶制备球孢白僵菌原生质体后,再生的最佳条件是:用L-broth培养基培养球孢白僵菌78h,KCl 0.8mol/L(再生用蔗糖0.6mol/L),柠檬酸-柠檬酸钠缓冲液为缓冲系统,纤维素酶:蜗牛酶:溶菌酶=5:2.5:2.5(mg/mL),在32℃水浴中酶解3h(pH自然)。

Abstract: The protoplast was prepared from mycelia of Beauveria bassiana(Balsamo) Vuillemin by enzymatic hydrolysis through liquid culture, and the conditions of regeneration were studied by means of one way experiment.The results showed that the optimal regeneration conditions were as follows:mycelium was cultured 78 h on L-broth culture medium, 0.8 mol/L KCl(regeneration:0.6 mol/L sucrose) was osmotic stabilizer, the citric acid sodium citrate was buffer system, Cellulase:Snailase:Lysozyme=5:2.5:2.5(mg/mL), with digesting 3 h in 32℃ water baths(natural pH).
