家兔PIK3CAAKT3基因多态性及其与生长性状的关联性分析

Polymorphisms of PIK3CA and AKT3 Genes and Their Association with Growth Traits of Rabbits

摘要: [目的] 探讨家兔PIK3CA和AKT3基因的多态性及其与家兔生长性状的相关性。[方法] 采用PCR产物直接测序法寻找PIK3CA和AKT3基因的遗传变异,应用PCR-RFLP技术对欧洲大白兔、伊拉兔和福建黑兔3个肉兔品种共711个个体进行基因型分析。[结果] 在PIK3CA和AKT3基因编码区发现了2个同义突变,分别是PIK3CA基因的c.3068 A>G,和AKT3基因的c.1213 A>C。对于c.3068 A>G,在欧洲大白兔和福建黑兔中,GG基因型个体的体重在各生长阶段均显著低于AA和AG基因型个体(P<0.05);对于c.1213 A>C位点,CC基因型的欧洲大白兔体重显著低于AA和AC基因型个体(P<0.05);综合考虑两位点,发现GGCC基因型的欧洲大白兔个体重极显著低于AAAA,AGAA和AGAC基因型个体。[结论] PIK3CA和AKT3基因与家兔的生长性能相关,可以作为家兔分子育种的遗传标记。

Abstract: [Objective] This study was to investigate the polymorphisms of PIK3CA and AKT3 Genes and their association with growth traits of rabbits. [Methods] We use PCR direct sequencing to detect the polymorphisms of PIK3CA and AKT3 genes and PCR-RFLP was applied to the genotyping analysis of 711 individuals including 3 meat rabbit breeds. [Results] A total of six SNPs of PIK3CA and two SNPs of AKT3 were finally found on the genomic sequences. And there were two synonymous mutations of their coding region, one was in the exon20 of PIK3CA(c.3068 A>G) and one was in exon10 of AKT3(c.1213 A>C), respectively. As for c.3068 A>G of PIK3CA, GG genotype individuals had the lowest weight at all growth stages in Fujian black rabbits and European white rabbits. The c.1213 A>C of AKT3 only exists in European white rabbits, and AC and AA genotype were significantly associated with higher body weight than CC genotype at every growth stage(P<0.05). The combined genotypes GGCC of c.3068 A>G and c.1213 A>C was associated with significantly lower body weight compared with other five combined genotypes in European white rabbits. We also found the breed-specific polymorphism, such as AA genotype individuals were not found in Ira rabbits at PIK3CA c.3068 A>G. [Conclusion] Our results indicated that the polymorphism of both PIK3CA and AKT3 genes were significantly associated with production traits in rabbits, which would be employed in the breeding program of marker assisted selection.
