基于EST-SSR的陕西茶树资源遗传多样性分析

Genetic Diversity Analysis of Tea in Shaanxi Province Based on SET-SSR

摘要: [目的] 明确陕西省主要茶树种质资源的遗传多样性,为茶树育种提供依据。[方法] 用6份有代表性的陕西茶树资源材料,从154对EST-SSR引物中筛选出39对扩增条带清晰、多态性高的引物,采用NTSYS-pc2.10e分析软件,相似系数选择DICE,用UPGMA进行聚类,对陕西省46份茶树资源进行遗传多样性研究。[结果] 共检测到316个等位位点,每对引物可检测到3~16个等位位点,平均为8个;每个EST-SSR位点的遗传多态性信息含量在0.76~0.99之间,平均值为0.94。供试材料的遗传相似系数在0.00~0.89之间。[结论] EST-SSR标记能够揭示材料间较高的遗传多样性,基于EST-SSR标记的陕西省主要茶树资源遗传多样性处于较高水平。

Abstract: [Objective] This study aimed to define the genetic diversity of tea germplasm in Shaanxi Province, and to provide the scientific basis for tea breeding. [Method] In this study, 6 representative tea plants were taken as templates and 39 pairs of EST-SSR markers were chosen from total 154 pairs of primers. The genetic polymorphism of 46 tea materials in Shaanxi Province was explored by NTSYS-pc2.10e. The similarity coefficient was DICE and UPGMA was used for clustering. [Result] Total 316 alleles were verified using EST-SSR markers. 3-16 alleles were detected by each pair of molecular markers, with an average of 8 alleles. The genetic polymorphism information content of each EST-SSRlocus ranged from 0.76 to 0.99, with an average of 0.94. Additionally, the genetic similarity of the 46 tea materials varied from 0.00 to 0.89. [Conclusion] The results showed that EST-SSR markers may reveale higher genetic polymorphism among the tested tea materials. Based on EST-SSR markers, the genetic diversity of these tea materials was relatively high in this study.
