葛渣异黄酮的提取工艺优化及抗氧化性研究

Extraction and Antioxidant Property of Isoflavones from Pueraria Root Residue

摘要: [目的] 优化葛渣异黄酮提取工艺并对其生物活性进行初步研究。[方法] 在单因素试验基础上,以异黄酮提取率为指标,采用响应面分析法研究乙醇浓度、料液比、提取温度和提取时间对异黄酮得率的影响,并建立回归模型。同时采用邻苯三酚自氧化法和水杨酸比色法测定葛渣异黄酮的抗氧化性。[结果] 在乙醇浓度70%,料液比1:17(g:mL),提取温度73℃,提取时间120 min的条件下,异黄酮提取率12.85 mg/g最高,与模型预测理论值差异不显著(P>0.05)。在0~180μg/m L浓度范围内,葛渣异黄酮对超氧阴离子自由基与羟基自由基的最大清除率分别为58.9%、82.0%,其IC50分别为170μg/m L、78μg/m L。[结论] 葛渣异黄酮具有较强的抗氧化作用,可用于天然抗氧化剂的开发。

Abstract: [Objective] Response surface methodology(RSM) was applied to optimize the extraction technology of isoflavones from pueraria root residue and the antioxidant activity of isoflavones was also studied in this paper. [Method] On the basis of single-factor test, the regression model was set up by RSM, involving ethanol concentration, material-liquid ratio, extraction temperature and extraction time as independent variables, and the extraction yield of isoflavone as the experimental index. The effect of removing free radicals of O2-· and ·OH of isoflavones was detected by pyrogallol autoxidation method and salicylic acid method respectively. [Results] Under the optimal extraction conditions of 70% ethanol, material-liquid ratio 1:17, extracting temperature 73℃ and extracting time 120 min, the yield of isoflavones could reach at 12.85 mg/g, which had no significantly difference with the predictive value(P>0.05). Isoflavones had strong scavenging effects on free radicals of O2-· and ·OH. In the range of 0-180 μg/mL, concentration of isoflavones was positively associated with scavenging rate. The scavenging activities to O2-· and ·OH were 58.9% and 82.0% at 180 μg/mL. Their IC50 values were 170 μg/mL and 78 μg/mL, respectively. [Conclusion] Isoflavones from pueraria root residue have better antioxidant effect and can be used as natural antioxidants.
