Spectrometric Study of the Interaction between Analgin and Bovine Serum Albumin and the Effect of Coexistent Metal Ions

摘要: [目的] 旨在研究安乃近(ANG)与牛血清白蛋白(BSA)的相互作用,为安乃近的临床研究提供一定的参考依据。[方法] 在优化的实验条件下,运用光谱法研究牛血清白蛋白(BSA)与安乃近(ANG)的相互作用及共存金属离子的影响。[结果] 研究表明:ANG对BSA的荧光有猝灭作用,属于静态猝灭。BSA能运输ANG,是一个自发过程,其作用力类型主要为氢键或范德华力。BSA的亚螺旋域ⅡA是主要结合位置,离酪氨酸残基更近,有微弱的药物负协同作用。ANG对BSA构象产生影响。Cr3+、Co2+、Fe3+、Mn2+和Mg2+对ANG与BSA结合有竞争作用,增强药效。[结论] ANG确实与BSA发生了相互作用,金属离子对两者结合产生影响。

Abstract: [Objective] The interaction of Analgin(ANG) with bovine serum albumin(BSA) was investigated so as to provide references for its clinical application. [Method] Under the optimal conditions, the interaction of ANG with BSA and the effect of coxisting metal ions were investigated by spectrometry. [Results] The results showed that the fluorescence of BSA was quenched by ANG. It was a static quenching process. BSA could transport ANG, which was spontaneous and mainly driven by Der Waals forces or hydrogen bond. The primary binding site for ANG was located at sub-domain ⅡA of BSA, which was near by tyrosine residue. There was some negative cooperative effect. The conjugation reaction could affect the conformation of BSA, which led to the polarity around BSA strengthened. Mn2+, Fe3+, Cr3+, Zn2+ and Mg2+ competed with the interaction, which could increasing medical effectiveness. [Conclusion] ANG can interact with BSA and the interaction was affected by metal ions.
