瓦氏黄颡鱼血液生理指标与外周血细胞组成及显微结构

Blood Physiological Indexes, Constitute and Microstructure of Peripheral Blood Cell in Pelteobagrus vachelli

摘要: [目的] 研究瓦氏黄颡鱼(Pelteobagrus vachelli)血液生理指标与外周血细胞组成及显微结构。[方法] 以血细胞计数法、氰化高铁血红蛋白法和浓度梯度法分别测定其血液生理指标中的血细胞数目、血红蛋白含量和红细胞脆性;以瑞士和吉姆萨联染法对外周血涂片染色,进行血细胞的分类计数、大小测量和显微观察。[结果] 瓦氏黄颡鱼红细胞数、白细胞数和血红蛋白含量分别是1.91×1012/L、4.19×109/L和60.89g/L;血细胞比积和血沉值分别是0.45L/L和20.64mm/h;刚开始溶血和完全溶血对应NaCl浓度为(0.45±0.01)%和(0.27±0.01)%,抗逆幅度是(0.18±0.01)%;血液涂片鉴定出红细胞、嗜中性粒细胞、淋巴细胞、单核细胞、血栓细胞等五种细胞,未观察到嗜酸性和嗜碱性粒细胞;各类血细胞体积由大到小排列顺序是红细胞、单核细胞、嗜中性粒细胞、大淋巴细胞、小淋巴细胞、血栓细胞;白细胞分类计数表明血栓细胞数目最多,其次是小淋巴细胞、嗜中性粒细胞、大淋巴细胞,单核细胞数目最少。

Abstract: [Objective] The research investigated blood physiological indexes, and constitute and microstructure of peripheral blood cell in Pelteobagrus vachelli. [Method] The blood physiological indexes were determined by count, cyanmethemoglobin determination and concentration gradient methods including red and white blood cells number, hemoglobin content and erythrocyte osmotic fragility. The differential leucocytes were counted, and the sizes of blood cells were measured through observing the peripheral blood cells smears which were stained by Wright's and Gimsa's stain. [Results] The results indicated that the number of red blood cells, white blood cells and hemoglobin content were 1.91×1012/L, 4.19×109/L, and 60.89g/L. The total number of erythrocytes was significantly higher than white blood cells;hematocrit and erythrocyte sedimentation rate were 0.45L/L and 20.64mm/h;the minimum and maximum erythrocyte osmotic fragility were (0.45±0.01)% and (0.27±0.01)%, amplitude for resistance was (0.18±0.01)%. On the stained smears, five major cell types were recognized:erythrocytes, neutrophils, lymphocytes, monocytes, and thrombocytes;some immature blood cells and direct dividing of erythrocytes were also be observed, while eosinophils and basophils were not found;among these cells, the sizes of blood cell was:erythrocytes > monocytes > neutrophils > large lymphocytes > small lymphocytes > thrombocytes;differential leucocyte counts showed that the thrombocytes were the most abundant cell type after erythrocytes followed by small lymphocytes, neutrophils, large lymphocytes, and monocytes.
