产弹性蛋白酶芽孢杆菌的筛选与鉴定

Screening and Identification of Elastase-producing Bacillus Strains

摘要: 从屠宰场,牛奶场和啤酒厂附近采集的土样和污水中,通过弹性蛋白平板筛选获得209株产弹性蛋白酶的细菌菌株,将弹性蛋白水解圈和菌落直径之比(HC)比值较大的83株菌通过摇瓶复筛后得到一株酶活力较高的菌株SCEL01,产酶活性达到33 u/mL。结合形态学、生理生化特性和16SrDNA序列同源性分析,该菌株被鉴定为枯草芽孢杆菌(Bacillus subtilis)。

Abstract: From the soil and wastewater samples near the slaughterhouse, milk factory and beer factory, 209 strains with elastase activity were isolated. Furthermore, 83 strains were collected because of the higher ratio of hydrolyze loop and colony diameter (HC). Finally, one strain with the highest elastase activity was screened and named strain SCEL01. Its elastase activity reached to 33 u/m L. The identification show ed that strain SCEL01 was Bacillus subtilis by the way of morphology, biochemistry characteristic and 16SrDNA sequence homology analysis.
