乌头种质资源遗传多样性的RAPD分析

The Genetic Diversity of Germplasms of Aconitum carmichaeli Debx. Revealed By RAPD Markers

摘要: 为了探讨乌头(Aconitum carmichaeli Debx.)种质资源的遗传多样性和亲缘关系,我们收集了来自于云南和四川的23份种质资源,采用2%CTAB法提取乌头总基因组DNA,用15个RAPD随机引物进行分析。15个引物共扩增出242个DNA片段,多态性片段202个,占总扩增片段比率为83.5%,平均每个引物可扩增出13.47个多态性DNA片段。供试材料的遗传相似系数在0.155~0.940之间,表明乌头种质资源具有丰富的遗传多样性。在遗传相似系数0.582处将23份供试材料划分为9个类群,其中15份聚为一个大类,其余8份各自聚为一类。RAPD分析可作为构建乌头DNA指纹图谱的有效方法。

Abstract: To analyse genetic diversity and sibling relationship among Aconitum carmichaeli Debx., 23 germplasms from Sichuan and Yunnan were sampled.Total genomic DNA were extracted from sampled materials by improved CTAB method, and RAPD markers were amplified by 15 random primers.Total 242 DNA bands with 202 polymorphic bands were amplified.An average of 13.47 polymorphic bands were amplified by each primer.The similarity coefficients ranged from 0.155~0.940, which revealed that different materials of Aconitum carmichaeli Debx.had rich genetic diversity.At the similarity coefficients of 0.582 the 23 germplasms were classified into 9 groups by clustering analysis.RAPD markers could be one effective method to construct DNA fingerprintings for Aconitum carmichaeli Debx.
