Genetic Variations of Storage Protein Genes among Chinese Endemic Wheats Detected by PCR Amplification

摘要: 利用Glu-A1xGlu-B1xGlu-A3、Glu-B3和Glu-1Dx5的特异性PCR引物,和位于1BS染色体上的γ-醇溶蛋白和低分子谷蛋白2对SSR标记,通过PCR的方法研究了8份四川白麦子、14份云南铁壳麦、9份西藏半野生小麦和9份新疆稻麦贮藏蛋白基因的遗传多样性。结果表明,γ-醇溶蛋白和低分子谷蛋白2个SSR位点的遗传多样性较高,其次为Glu-A3和Glu-B3,而Glu-AxGlu-Bx的遗传多样性最低。在所有40份供试材料均未扩增出Glu-1Dx5基因的特异DNA片段,说明这些小麦地方品种不含优质亚基5的编码基因。

Abstract: Five special PCR primers for Glu-A1x, Glu-B1x, Glu-A3, Glu-B3 and Glu-1Dx5, and 2 SSR primers for γ-gliadin and LMW-glutenin were used to investigate the genetic variations of storage protein genes among 40 Chinese endemic wheat accessions, including 8 Sichuan white wheat landraces, 14 Yunnan hulled wheat accessions, 9 Tibetan weedrace and 9 Xinjiang rice wheat accessions. The PCR analysis indicates that low level of genetic diversity is found at Glu-Ax, Glu-Bx, Glu-A3 and Glu-B3, while high level of genetic diversity is founded at the SSR loci of γ-gliadin and LMW-glutenin. The special DNA fragment of Glu-1Dx5 is not detected among all 40 Chinese endemic wheat accessions. It indicates that there is no subunit 5, encoded by Glu-D1 among these accessions.
