中江大叶丹参离体培养技术研究

A Study on Vitro Culture Technique of Zhongjiang Large-leaf Type Salvia

摘要: 实验以中江大叶丹参初展开叶片为外植体研究丹参快繁技术。结果表明:初代培养以1/2MS+3%蔗糖+0.7%琼脂+3mg/L6-BA培养基为佳,在此培养基上培养60d后可获得63.3%的芽苗分化率;继代增殖培养以2/3MS+3%蔗糖+0.7%琼脂+0.4mg/L6-BA培养基为优,在此培养基上每4周可获得5.4倍的增殖率;无根试管苗可采用瓶外生根法生根,用50mg/LIBA和50mg/LNAA混合液浸泡无根试管苗基部5min,以疏松肥沃壤土为基质,置于阴凉湿润塑料大棚内,6周后可获得86%的生根率。

Abstract: A micropropagation technique of salvia (Salvia miltiorrhiza Bge) was developed by using expanding young leaves for explants in this study.A shoot induction rate of 63.3% can be obtained 60 days after inoculation on the medium containing 1/2 MS+3% sucrose+0.7% agar+0.4 mg/L 6-BA,resulting in a multiplication rate of 5.4 every four weeks.Rooting the shoots ex vitro results in a root formation rate of 86% in 6 weeks by treating the base of the shoots for 5 minutes in 50 mg/L IBA and 50 mg/L NAA solution and then culturing them in a well aerated fertile soil medium.
