枇杷胚乳愈伤组织诱导和不定芽发生的研究

摘要: 以枇杷人工授粉5~8周的幼果为材料,取细胞形成期的胚乳进行培养,结果表明:低温处理的幼果的胚乳形成愈伤组织的频率有所提高;来自预培养在MS分别附加6-BA,GA3,2,4-D的3种培养基中2~4d的胚乳愈伤组织诱导频率明显提高;6-BA、2,4-D、GA3对愈伤组织诱导频率有显著影响,其中2,4-D是诱导胚乳愈伤组织的主导因素,在3种激素的共同作用下,诱导率最高达33.3%。经1~2代增殖的愈伤组织在培养基MS+ZT2mg/L+NAA 0.1mg/L和MS+ZT 3mg/L+NAA 0.1mg/L中分化芽苗数最多,芽苗分化率最高达18.04%。

Abstract: Endosperm at the stage of cell formation, excised from artificially pollinated fruit of loquat, was suitable for culture in vitro. The result indicated that endosperm tissues, excised from the fruits treated with low temperature(4~7 ℃) for 17 days, and from the immature seeds precultured in MS basal medium added respectively with 6-BA, 2, 4-D, GA3 for 2~4 days, stimulated callus formation; 6-BA or 2, 4-D or GA3 had much influence on the frequency of callus induction, with 2, 4-D being a leading factor among them. As a result of the combined effect of 6-BA, 2, 4-D, GA3, the percentage of callus induction was as high as 33.3%. Endosperm callus, cultured in medium MS+ZT 3 mg/L+NAA 0.1 mg/L and MS+ZT 2 mg/L+NAA 0.1 mg/L, could regenerate more shoots than in any other media, with the percentage of shoots regenerated being as high as 18.04%.
