四川省部分小麦新品系醇溶蛋白遗传多样性分析

Gliandin Analysis of the Genetic Diversity in a few of Common Wheat Lines Grown in Sichuan, China

摘要: 利用酸性聚丙烯酰胺凝胶电泳(APAGE)对2001年参加四川省区试的20个小麦新品系和2个对照品种进行了醇溶蛋白基因位点的特异性检测,分析了不同品系(种)间醇溶蛋白带型的遗传差异。结果表明,22个小麦品系(种)具有22种带型,每个材料分离出18~29条带,22个材料共分离出56条带,其中49条具有多态性,占87.5%。20个新品系中具有1RS/1BL易位系标记性位点Gli-B1l的有11个,占供试新品系的55%。22个材料间的遗传距离在0.043~1.00间,平均值为0.444;1RS/1BL易位系与非1RS/1BL易位系间的遗传距离较大。基于遗传距离的聚类分析表明:供试材料在遗传距离0.50水平上明显聚为4类。分析表明,供试四川小麦新品系具有较为广泛的基于醇溶蛋白带谱的遗传多样性。同时还探讨了供试材料中Gli-B1l位点高频率存在的原因及进一步合理利用1B/1R易位系的可能性。

Abstract: Gliadin variation of 20 Sichuan wheat lines having entered the province key contest and 2 common wheat cultivars as contrast were investigated by acid ployacrlamide gel electrophoresis. The results are as follows:there are 22 gliadin patterns in the 22 materials, with 9 lines bearing the allele Gli-B1l, a marker of 1RS/1BL translocation chromosome. 56 gliadin bands are separated by electrophoresis and 49 out of them show polymorphism(amount to 87.5%). The genetic distance(GD) based on gliadin patterns ranges from 0.043 to 1.00, with a mean value of 0.444. There are relatively higher GD between the 1RS/1BL and non 1RS/1BL translocation lines. Cluster analysis shows that the studied materials can be classified into 4 clusters at the level of GD 0.50. Based on these results, it is concluded that the studied common wheat lines have respectively higher genetic diversity. The reason why there is a high frequency of the allele Gli-B1l and the potentiality of reasonably utilizing 1BL/1RS wheat translocation lines is also discussed in the paper.
